I apply for the Tax graduate programme, and wondering when would we know what exact tax line we will be allocated into.

For instance, corporate tax, direct tax, VAT etc.

Thanks for your question - you're told roughly before your EY Experience Day if you have an interview as it will be for that team, and the student recruitment team will let you know in advance of that day to ensure you have time to prep and do any additional research into the specific business area.
